He was part of the cast in the mini-series film, On the Job: The Missing 8 (2021-2022) directed by Erik Matti. The movie was internationally acclaimed and was nominated at the 50th International Emmy Awards for Best TV Movie or Mini-series. The cast won for Best Ensemble Performance by the Society of Filipino Film Reviewers. The series was also selected as the Philippines entry to Best Feature Film at the 95th Academy Awards.

In 2024, he top billed the prison drama movie, Green Bones directed by Zig Dulay. He played the role of Domingo Zamora, a criminal accused of murdering his family. His acting in the movie won him several Best Actor Awards in local and international award giving bodies including Best Actor for Gawad Urian Award, Best Actor award at the 50th Metro Manila Film Festival, Best Actor Award for Gawad Pasado Award, Asian Academy Creative Award for Best Actor in a Leading Role in the national category and Best Actor Award in the International Golden Summit Excellence Award in Vietnam as well as received a nomination from FAMAS Award.

On television, he starred in the drama series Magdusa Ka (2008) directed by Maryo J. Delos Reyes. The series was nominated for Best Teleserye at the 37th International Emmy Awards. In 2013, Trillo lead the cast in the drama series, My Husband’s Lover directed by Dominic Zapata playing the role of Eric, a gay lover of a married man.[5] His role won him Best Actor award at the Asian Television Awards, Best Actor at the Golden Screen TV Awards, Best Actor at Gawad Tanglaw Award and a nomination from PMPC Star Awards for Television. The series was nominated at the 42nd International Emmy Awards.

He starred with Jennylyn Mercado, his wife in the drama series, My Faithful Husband directed by Joyce Bernal. His role as Emmanuel won him an Asian Star Prize Award in 2016 Seoul International Drama Awards as well as a nomination from PMPC Star Awards for Television.[9]

In 2024, he played as the main antagonist in the war drama series, Pulang Araw directed by Dominic Zapata. He won Best Supporting Actor award at the PMPC Star Awards for Television for playing the role of Yuta Saitoh, a Japanese Imperial Officer in the series. The series was nominated for Best Telenovela at the Venice TV Awards in Italy.
